Yoon Eun Hye memulai debutnya sebagai penyanyi, ia bergabung dalam sebuah grup vokal terkenal bernama Baby VOX sejak tahun 1999 sampai 2005. Pada tahun 2005 Baby VOX secara resmi dibubarkan dan Yoon Eun Hye memilih untuk bersolo karir, hingga akhirnya merambah ke dunia akting. Ia muncul sebagai anggota reguler dari sebuah acara variety show terkenal Xman.
Yoon Eun Hye memperoleh kesempatan besar ketika berperan sebagai pemimpin wanita dalam drama Goong (Princess Hours). Goong menjadikan Yoon Eun Hye sebagai aktris terkenal, baik di Korea Selatan maupun di luar negeri. Drama berikutnya setelah Goong adalah sebuah drama komedi berjudul The Vineyard Man. Drama ketiganya berjudul Coffee Prince, drama tersebut sangat terkenal dan menempaatkan Yoon Eun Hye sebagai superstar urutan A di industri hiburan Korea sekaligus juga sebagai bintang hallyu di luar negeri.
Yoon Eun Hye kembali muncul dilayar televisi pada 2009 dengan drama My Fair Lady, drama tersebut merupakan drama kemunculan kembali Yoon Eun Hye setelah lebih dari 2 tahun aejak drama Coffee Prince.
